Sorry, I'm afraid I don't know. It probably isn't from the line of William Spencer (grandfather of Anne Sheppard), because according to "Adventures of Purse and Person" his descendants are through his daughters. So the name "Spencer" died out in that line way back in the 17th century.

However, there are many Spencer lines besides that one. The Spencer DNA study lists a lot of lines at http://www.familytreedna.com/public/SPENCER -- you've probably already looked at that, but in case you haven't, it might be worth having a look to see if anything there rings any bells.
